Melanotan 2 is an innovative way to stimulate tanning and prevent the harmful effects of UV radiation. MT2 is a synthetic peptide analogue of the natural hormone malenocortin, which is the main factor that stimulates skin cells to produce melanin, the pigment that colors the skin and protects it from harmful UV rays. Melanotan 2 has a number of effects that are so attractive to people of different ages and genders: stimulation and increase in sexual desire (increased libido functions), and also reduces appetite, thanks to which it is possible to get rid of excess weight.
There are two types of this peptide: Melanotan 1 and Melanotan 2. The first – Melanotan 1 is very gentle, with systematic effects, which is why it has already been approved for use in Italy. Melanotan 2 surpasses Melanotan 1 several times in terms of its effect on melanocortin receptors, which makes it more effective for quick tanning, but Melanotan 2’s tan is more pronounced.
Mechanism of action
Melanotan 2 is a hypothalamic hormone that belongs to the melanotropin family, a group of peptides that react with melanocortin receptors and has a wide range of effects on almost all types of tissues. Other melanotropic hormones include ACTH and alpha melanocyte stimulating hormone (a-MSH). Melanotan 2 is an alpha-melanocyte-stimulating hormone analog structurally modified to reduce enzymatic degradation and increase receptor binding. Initially, the properties of the hormone to cause sunburn and pigmentation were studied, but more recent studies have focused on its antioxidant properties, the effect on the shift in physiological rhythms and the possible use of the drug as a therapy for erectile dysfunction. Melanotan 2 is a broad and powerful stimulant of the melanocortin system.
Melanocortin receptors are the main mechanism through which Melanotan 2 exerts its action. MCR or melanocortin receptors exist in various functions in the human body, as well as in many other organisms. All MCRs consisting of various subtypes that perform various functions constitute the melanocortin system. The melanocortin system performs various functions in the body using different mechanisms based on a limited number of functional groups – it has profound systemic effects in central and peripheral interactions with other known neuropeptide systems (leptin, ghelin, insulin, etc.)
These functions include control of energy balance, thyroid function and insulin response. Furthermore, MCRs play an important role in skin pigmentation and also influence reproductive function on both physical and behavioral levels. When combined with endogenous opioids, they are able to modulate pain and nerve sensitivity, respond to inflammation, as well as exert neuroprotective effects (including resistance to oxidative stress), influence aggression, and modulate circadian rhythms, including excitement while waiting for food.
Effects of pigmentation
Pigmentation Effects Melanotan 2 was initially studied for its effect in including skin pigmentation mediated by MK-1R receptor activity. The accidental discovery that Melanotan 2 also induces erections and increases libido in both men and women led to a larger study of Melanotan 2 for the treatment of sexual dysfunction, however, so far pigmentation is perhaps the best known quality of Melanotan 2 Melanotan 2 is said to be able to provide “therapeutic tanning” even in people at risk: the expression of melanin causes dark skin pigmentation associated with a reduced risk of various skin cancers and UV damage.
Improving effects on erection and libido
In men and women Melanotan 2 is able to cause acute sexual arousal, an increase in libido and (in men) an increase in erection parameters. Melanotan 2 is effective in treating psychogenic erectile dysfunction and other types of erectile dysfunction. The erection-enhancing properties of melanotan 2 are not limited to cases of psychogenic erectile dysfunction. Penile erection has been observed in men with various organic factors. The findings of increased sexual desire warrant further investigation of the effects of these centrally acting drugs on sexual desire disorders. Since Melanotan 2 works mainly in the brain, it is more effective in treating arousal disorders and causes genital arousal and increases libido in both women and men. However, various researchers have found that the erectile properties of Melanotan 2 are partly responsible for the neuronal release of nitric oxide.
Melanocortin system and obesity
The melanocortin system is a potential therapeutic target for reducing appetite and weight, increasing fatty acid oxidation, and treating obesity-associated diseases such as insulin resistance, diabetes, and hypertension. Alpha-melanocyte stimulating hormone, the analogue of which is Melanotan 2, is the main endogenous stimulator of the human melanocortin system. The alpha-MSH response in obesity demonstrated by animal studies is a good indication of the potential of alpha-MSH or analogues such as Melanotan 2 in the treatment of obesity by reducing weight, restoring central and peripheral sensitivity to other hormones (such as thyroid hormones) and reduce hunger. Melanotan 2 significantly reduced calorie intake in rats (-32+/-2 and -25+/-2 kcal/day), increased insulin sensitivity, and improved glucose tolerance. This is also a meta-analysis that supports some weight loss in humans.
Melanocortin system and inflammation
Receptors of the melanocortin system play a role in models of experimental inflammation. The study reports that Malanotan 2 has anti-inflammatory effects and may improve blood vessel health.

How to use
Do not use Melanotan 2 in large doses and with fanatical consistency. In the first week, do not abuse ultraviolet and do not forget to moisturize the skin. Fairly normal dosages for people with white skin will be 100-200 mcg per day or every day.
To achieve the desired skin tone and minimize the likelihood of “melanotic freckles”, do not exceed the dosages and do not stay in the sun for a long time. Let your body store melanin on its own. Usually the drug is used for 2-3 weeks, until the desired skin tone is achieved, then a reduced dose of two times or less is taken to maintain the tan. The recommendation for the first injection is to avoid side effects in the form of tachycardia and redness of the skin and to take 50-100 mcg.
One case of overdose has been recorded, i.e. administration of 6 mg once. Some time after the injection, we felt severe pain that spread to the entire body. Doctors found a significant increase in heart rate and increased blood pressure; Urinalysis showed a significant increase in the release of myoglobin, a protein that is an indicator of muscle damage. A high dose of Melanotan 2 caused the destruction of muscle fibers, resulting in increased plasma proteins and, consequently, kidney damage.
Further studies have shown that Melanotan 2 can be potentially harmful to muscles (and the body as a whole) at dosages above 6 mg per day. In any case, it is not recommended to exceed the daily dose of 1 mg, and if you feel heart disease or pain throughout the body, taking the drug should be stopped immediately.
How to prepare a solution
To prepare an injectable solution, you take a syringe already containing a diluent and inject it into a vial containing a lyophilized powder. Tilt the vial so that the needle touches the wall of the vial. Avoid injecting the diluent directly into the lyophilized powder. The solvent should flow slowly down the wall of the bottle (don’t fill it all at once and take your time). Once all the diluent has been added to the peptide vial, mix gently (but do not shake or shake the vial) until the lyophilized powder has dissolved and you have a clear liquid. Now the drug is ready for use.
Never mix one peptide with another in the same syringe. This creates the risk that the fragile peptide molecules will be destroyed.
Application
- The injection can be subcutaneous, intramuscular, depending on personal preferences.
Archiving
- The resulting solution can be stored for approximately 21 days in the refrigerator at a temperature of 2-8 degrees Celsius. The storage time increases if the solution was prepared using bacteriostatic water.
